PCI-24781 â CHECKS THE GROWTH OF CANCER VIA A NOVEL MECHANISM

INTRODUCTION

Epigenetics that is alterations in gene expressions, accompanied with unaltered DNA sequence is also being considered as one of the primary cause of cancer. These alterations can be due to various reasons like methylation of DNA by methyl transferases, modifications of histone proteins due to HATs and HDACs. Aberrations in histone modifications are related with silencing of the tumor suppressor genes. This leads to cancer. HDAC inhibitors target these modifications of the histone proteins and the act against the HDAC enzymes.

PCI-24781: ITS CYTOTOXIC EFFECTS

Within mammals 11 HDAC enzymes have been discovered which have been categorized into four different classes. The classification of HDAC inhibitors is also done based upon the class of HDACs that they inhibit. PCI-24781 was previously known as CRA-024781 and belongs to the class of phenyl hydroxamic acid derived compounds. From the compound library of HDAC inhibitors, PCI-24781 has been specifically selected for its action against action against all isoforms of HDACs belonging to class I and II. At a low concentration it shows inhibitory effect against HDACs belonging to class I and III, with a greater potency. Under in vitro conditions it showed inhibitory effect against various cancer cell lines. Apoptosis and histone alterations due to PCI-24781 are closely associated with caspase-8 and FADD (Fas-associated death domain). Especially in case of acute leukemia, PCI-24781 stimulates the raise in superoxide levels. The raise in reactive oxygen species lies downstream to the action of PCI-24781 on histone H3 proteins. Hence this research helped in analyzing the roles of caspase-8, FADD and ROS within PCI-24781 stimulated cells [1].

PCI-24781: CHECKS HOMOLOGOUS RECOMBINATION

PCI-24781 inhibits the DNA repair mechanism within cancer cells. The double stranded breaks introduced during the normal course are repaired through recombinational repair of the homologous chromososmes. PCI-24781 acted synergistically along with PJ34 and stimulated apoptosis within cancerous cells. PCI-24781 completely inhibited the repair mechanism within the subnuclear foci region. The inhibition by this agent halts the transcription of those genes which are responsible for homologous recombination for example RAD51 [2].

PCI-24781: STIMULATES APOPTOSIS THROUGH NF-ΚB MECHANISM

PCI-24781 was found to be synergistic with Bortezomib in terms of cytotoxicity and induction of cell death. It arrests the cell cycle at the GAP0/GAP1 phases. It diminished the synthetic phase and stimulated the expression of p21 protein. It also promoted the synthesis of reactive oxygen species within lymphoma cell lines. Together with bortezomib, it stimulates the cleavage of caspase, depolarization of mitochondria and acetylation of histone proteins. These effects were either not visible or seen at a lower intensity when both the agents were introduced alone. The genes required for NF-κB pathway were down regulated. This suggests that PCI-24781 stimulates apoptosis through NF-κB inhibition and caspase dependent mechanism [3]. Research has shown that this inhibitor is also effective against cancers associated with soft tissues. Within these cells it stimulates apoptosis, arrests the cell cycle in the GAP2 phase and depletes the cells within the synthetic phase. It works very efficiently in combination with chemotherapeutic agents by inhibiting the HRR (homologous recombination repair) [4].

CONCLUSION

In summary PCI-24781 acts as a potent anti-cancerous agent by controlling the DNA repair mechanism within the cancerous cells. It shows coordinated and superior effects with other chemotherapeutic agents.
