PCI-24781 – CHECKS THE GROWTH OF CANCER VIA A NOVEL MECHANISM

INTRODUCTION

Epigenetics that is alterations in gene expressions, accompanied with unaltered DNA sequence is also being considered as one of the primary cause of cancer. PCI-24781: ITS CYTOTOXIC EFFECTS

PCI-24781 was previously known as CRA-024781 and belongs to the class of phenyl hydroxamic acid derived compounds. The inhibition by this agent halts the transcription of those genes which are responsible for homologous recombination for example RAD51 [2].

PCI-24781: STIMULATES APOPTOSIS THROUGH NF-ΚB MECHANISM

PCI-24781 was found to be synergistic with Bortezomib in terms of cytotoxicity and induction of cell death. It arrests the cell cycle at the GAP0/GAP1 phases. It diminished the synthetic phase and stimulated the expression of p21 protein. It also promoted the synthesis of reactive oxygen species within lymphoma cell lines. Together with bortezomib, it stimulates the cleavage of caspase, depolarization of mitochondria and acetylation of histone proteins. These effects were either not visible or seen at a lower intensity when both the agents were introduced alone. The genes required for NF-κB pathway were down regulated. This suggests that PCI-24781 stimulates apoptosis through NF-κB inhibition and caspase dependent mechanism [3]. Research has shown that this inhibitor is also effective against cancers associated with soft tissues. Within these cells it stimulates apoptosis, arrests the cell cycle in the GAP2 phase and depletes the cells within the synthetic phase. It works very efficiently in combination with chemotherapeutic agents by inhibiting the HRR (homologous recombination repair) [4].

CONCLUSION

In summary PCI-24781 acts as a potent anti-cancerous agent by controlling the DNA repair mechanism within the cancerous cells. It shows coordinated and superior effects with other chemotherapeutic agents.
